The amphioxus ER acts as a dominant negative estrogen receptor. A pSG5 construct containing human ERα (A) or human ERβ (B) was tested in transfected HeLa cells for its ability to activate the co-transfected cognate ERE-luc reporter plasmid after E2 stimulation (10-6M) in presence of increasing doses of the amphiER construct. (C) A pSG5 construct containing human ERβ was tested in transfected HeLa cells for its ability to activate the co-transfected ps2 promoter after E2 stimulation (10-6M) in the presence of increasing doses of the amphiER construct.
