Regulation of β-catenin activity by Amer proteins. Co-transfection experiments of β-catenin and mouse Amer expression constructs together with a luciferase reporter gene (TOP-flash) into HEK293T cells are shown. The TOP-FLASH construct is designed to measure transcriptional activation mediated by β-catenin. As previously reported, WTX/AMER1 acts as a negative regulator of β-catenin activity. In contrast, AMER2 and AMER3 do not interfere with β-catenin dependent activation of the TOP-flash reporter gene. Luciferase values were normalized to β-galactosidase.
